Extensive research has elucidated the signaling mechanisms governing ERK activation via biochemical regulations with upstream molecules, particularly receptor tyrosine kinases (RTKs). However, recent advances have highlighted the role of mechanical forces in activating the RTK–ERK signaling pathways, thereby opening new avenues of research into mechanochemical interplay in multicellular tissues. Here, we review the force-induced ERK activation in cells and propose possible mechanosensing mechanisms underlying the mechanoresponsive ERK activation. We conclude that mechanical forces are not merely passive factors shaping cells and tissues but also active regulators of cellular signaling pathways controlling collective cell behaviors."}],"author":[{"last_name":"Hirashima","full_name":"Hirashima, Tsuyoshi","first_name":"Tsuyoshi"},{"first_name":"Naoya","id":"5299a9ce-7679-11eb-a7bc-d1e62b936307","full_name":"Hino, Naoya","last_name":"Hino"},{"first_name":"Kazuhiro","last_name":"Aoki","full_name":"Aoki, Kazuhiro"},{"full_name":"Matsuda, Michiyuki","last_name":"Matsuda","first_name":"Michiyuki"}],"department":[{"_id":"CaHe"}],"article_type":"review","fulldoi":"https://doi.org/10.1016/j.ceb.2023.102217","has_accepted_license":"1","day":"01","oa_version":"Published Version","tmp":{"legal_code_url":"https://creativecommons.org/licenses/by/4.0/legalcode","name":"Creative Commons Attribution 4.0 International Public License (CC-BY 4.0)","short":"CC BY (4.0)","image":"/images/cc_by.png"},"publication_identifier":{"issn":["0955-0674"],"eissn":["1879-0410"]}},{"day":"01","publication_identifier":{"eissn":["1879-0410"],"issn":["0955-0674"]},"oa_version":"None","article_type":"original","department":[{"_id":"EdHa"}],"author":[{"id":"3A9DB764-F248-11E8-B48F-1D18A9856A87","first_name":"Edouard B","orcid":"0000-0001-6005-1561","last_name":"Hannezo","full_name":"Hannezo, Edouard B"},{"last_name":"Simons","full_name":"Simons, Benjamin D.","first_name":"Benjamin D."}],"fulldoi":"https://doi.org/10.1016/j.ceb.2019.04.008","month":"10","abstract":[{"lang":"eng","text":"Branching morphogenesis is a prototypical example of complex three-dimensional organ sculpting, required in multiple developmental settings to maximize the area of exchange surfaces. It requires, in particular, the coordinated growth of different cell types together with complex patterning to lead to robust macroscopic outputs. In recent years, novel multiscale quantitative biology approaches, together with biophysical modelling, have begun to shed new light of this topic. Here, we wish to review some of these recent developments, highlighting the generic design principles that can be abstracted across different branched organs, as well as the implications for the broader fields of stem cell, developmental and systems biology."}],"publisher":"Elsevier","article_processing_charge":"No","year":"2019","publication_status":"published","status":"public","date_published":"2019-10-01T00:00:00Z","type":"journal_article","user_id":"ba8df636-2132-11f1-aed0-ed93e2281fdd","title":"Multiscale dynamics of branching morphogenesis","date_updated":"2026-04-03T09:38:46Z","scopus_import":"1","publication":"Current Opinion in Cell Biology","citation":{"mla":"Hannezo, Edouard B., and Benjamin D. Among these, the VCAM-1 receptor alpha4beta1 has received most attention as a main factor mediating firm adhesion to the endothelium during blood cell extravasation. Therapeutic trials are ongoing into the use of antibodies and small molecule inhibitors to target this interaction and hence obtain anti-inflammatory effects. However, extravasation is only one possible process that is mediated by beta1 integrins and there is evidence that they also mediate leukocyte retention and positioning in the tissue, lymphocyte activation and possibly migration within the interstitium. Genetic mouse models where integrins are selectively deleted on blood cells have been used to investigate these functions and further studies will be invaluable to critically evaluate therapeutic trials."}],"month":"10","fulldoi":"https://doi.org/10.1016/j.ceb.2006.08.007","author":[{"full_name":"Sixt, Michael K","last_name":"Sixt","orcid":"0000-0002-6620-9179","first_name":"Michael K","id":"41E9FBEA-F248-11E8-B48F-1D18A9856A87"},{"first_name":"Martina","last_name":"Bauer","full_name":"Bauer, Martina"},{"first_name":"Tim","last_name":"Lämmermann","full_name":"Lämmermann, Tim"},{"first_name":"Reinhard","full_name":"Fässler, Reinhard","last_name":"Fässler"}],"article_type":"original","oa_version":"None","publication_identifier":{"issn":["0955-0674"],"eissn":["1879-0410"]},"publist_id":"2191","day":"01","extern":"1"}]
