text: 'Transporters of the solute carrier 6 (SLC6) family translocate their cognate
substrate together with Na+ and Cl−. Detailed kinetic models exist for the transporters
of GABA (GAT1/SLC6A1) and the monoamines dopamine (DAT/SLC6A3) and serotonin (SERT/SLC6A4).
Here, we posited that the transport cycle of individual SLC6 transporters reflects
the physiological requirements they operate under. We tested this hypothesis by
analyzing the transport cycle of glycine transporter 1 (GlyT1/SLC6A9) and glycine
transporter 2 (GlyT2/SLC6A5). GlyT2 is the only SLC6 family member known to translocate
glycine, Na+, and Cl− in a 1:3:1 stoichiometry. We analyzed partial reactions
in real time by electrophysiological recordings. Contrary to monoamine transporters,
both GlyTs were found to have a high transport capacity driven by rapid return
of the empty transporter after release of Cl− on the intracellular side. Rapid
cycling of both GlyTs was further supported by highly cooperative binding of cosubstrate
ions and substrate such that their forward transport mode was maintained even
under conditions of elevated intracellular Na+ or Cl−. The most important differences
in the transport cycle of GlyT1 and GlyT2 arose from the kinetics of charge movement
and the resulting voltage-dependent rate-limiting reactions: the kinetics of GlyT1
were governed by transition of the substrate-bound transporter from outward- to
inward-facing conformations, whereas the kinetics of GlyT2 were governed by Na+
binding (or a related conformational change). Kinetic modeling showed that the
kinetics of GlyT1 are ideally suited for supplying the extracellular glycine levels
    required for NMDA receptor activation.'

text: Biochemical reactions often occur at low copy numbers but at once in crowded
and diverse environments. Space and stochasticity therefore play an essential
role in biochemical networks. Spatial-stochastic simulations have become a prominent
tool for understanding how stochasticity at the microscopic level influences the
macroscopic behavior of such systems. While particle-based models guarantee the
level of detail necessary to accurately describe the microscopic dynamics at very
low copy numbers, the algorithms used to simulate them typically imply trade-offs
between computational efficiency and biochemical accuracy. eGFRD (enhanced Green’s
Function Reaction Dynamics) is an exact algorithm that evades such trade-offs
by partitioning the N-particle system into M ≤ N analytically tractable one- and
two-particle systems; the analytical solutions (Green’s functions) then are used
to implement an event-driven particle-based scheme that allows particles to make
large jumps in time and space while retaining access to their state variables
at arbitrary simulation times. Here we present “eGFRD2,” a new eGFRD version that
implements the principle of eGFRD in all dimensions, thus enabling efficient particle-based
simulation of biochemical reaction-diffusion processes in the 3D cytoplasm, on
2D planes representing membranes, and on 1D elongated cylinders representative
of, e.g., cytoskeletal tracks or DNA; in 1D, it also incorporates convective motion
used to model active transport. We find that, for low particle densities, eGFRD2
is up to 6 orders of magnitude faster than conventional Brownian dynamics. We
exemplify the capabilities of eGFRD2 by simulating an idealized model of Pom1
gradient formation, which involves 3D diffusion, active transport on microtubules,
and autophosphorylation on the membrane, confirming recent experimental and theoretical
results on this system to hold under genuinely stochastic conditions.

text: "In two-player games on graphs, the players move a token through a graph to
produce an infinite path, which determines the qualitative winner or quantitative
payoff of the game. In bidding games, in each turn, we hold an auction between
the two players to determine which player moves the token. Bidding games have
largely been studied with concrete bidding mechanisms that are variants of a first-price
auction: in each turn both players simultaneously submit bids, the higher\r\nbidder
moves the token, and pays his bid to the lower bidder in Richman bidding, to the
bank in poorman bidding, and in taxman bidding, the bid is split between the other
player and the bank according to a predefined constant factor. Bidding games are
deterministic games. They have an intriguing connection with a fragment of stochastic
games called \r\n randomturn games. We study, for the first time, a combination
of bidding games with probabilistic behavior; namely, we study bidding games that
are played on Markov decision processes, where the players bid for the right to
choose the next action, which determines the probability distribution according
to which the next vertex is chosen. We study parity and meanpayoff bidding games
on MDPs and extend results from the deterministic bidding setting to the probabilistic
    one."

text: "Smart contracts are programs that are stored and executed on the Blockchain
and can receive, manage and transfer money (cryptocurrency units). Two important
problems regarding smart contracts are formal analysis and compiler optimization.
Formal analysis is extremely important, because smart contracts hold funds worth
billions of dollars and their code is immutable after deployment. Hence, an undetected
bug can cause significant financial losses. Compiler optimization is also crucial,
because every action of a smart contract has to be executed by every node in the
Blockchain network. Therefore, optimizations in compiling smart contracts can
lead to significant savings in computation, time and energy.\r\n\r\nTwo classical
approaches in program analysis and compiler optimization are intraprocedural and
interprocedural analysis. In intraprocedural analysis, each function is analyzed
separately, while interprocedural analysis considers the entire program. In both
cases, the analyses are usually reduced to graph problems over the control flow
graph (CFG) of the program. These graph problems are often computationally expensive.
Hence, there has been ample research on exploiting structural properties of CFGs
for efficient algorithms. One such well-studied property is the treewidth, which
is a measure of tree-likeness of graphs. It is known that intraprocedural CFGs
of structured programs have treewidth at most 6, whereas the interprocedural treewidth
cannot be bounded. This result has been used as a basis for many efficient intraprocedural
analyses.\r\n\r\nIn this paper, we explore the idea of exploiting the treewidth
of smart contracts for formal analysis and compiler optimization. First, similar
to classical programs, we show that the intraprocedural treewidth of structured
Solidity and Vyper smart contracts is at most 9. Second, for global analysis,
we prove that the interprocedural treewidth of structured smart contracts is bounded
by 10 and, in sharp contrast with classical programs, treewidth-based algorithms
can be easily applied for interprocedural analysis. Finally, we supplement our
theoretical results with experiments using a tool we implemented for computing
treewidth of smart contracts and show that the treewidth is much lower in practice.
We use 36,764 real-world Ethereum smart contracts as benchmarks and find that
they have an average treewidth of at most 3.35 for the intraprocedural case and
    3.65 for the interprocedural case.\r\n"

text: Biomarkers of aging can be used to assess the health of individuals and to
study aging and age-related diseases. We generate a large dataset of genome-wide
RNA-seq profiles of human dermal fibroblasts from 133 people aged 1 to 94 years
old to test whether signatures of aging are encoded within the transcriptome.
We develop an ensemble machine learning method that predicts age to a median error
of 4 years, outperforming previous methods used to predict age. The ensemble was
further validated by testing it on ten progeria patients, and our method is the
only one that predicts accelerated aging in these patients.

text: Distant luminous Lyman-α emitters (LAEs) are excellent targets for spectroscopic
observations of galaxies in the epoch of reionisation (EoR). We present deep high-resolution
(R = 5000) VLT/X-shooter observations, along with an extensive collection of photometric
data of COLA1, a proposed double peaked LAE at z = 6.6. We rule out the possibility
that COLA1’s emission line is an [OII] doublet at z = 1.475 on the basis of i)
the asymmetric red line-profile and flux ratio of the peaks (blue/red=0.31 ± 0.03)
and ii) an unphysical [OII]/Hα ratio ([OII]/Hα >  22). We show that COLA1’s observed
B-band flux is explained by a faint extended foreground LAE, for which we detect
Lyα and [OIII] at z = 2.142. We thus conclude that COLA1 is a real double-peaked
    LAE at z = 6.593, the first discovered at z >  6. COLA1 is UV luminous (M1500 = −21.6 ± 0.3),
has a high equivalent width (EW0,Lyα = 120−40+50 Å) and very compact Lyα emission
    (r50,Lyα = 0.33−0.04+0.07 kpc). Relatively weak inferred Hβ+[OIII] line-emission
from Spitzer/IRAC indicates an extremely low metallicity of Z <  1/20 Z⊙ or reduced
strength of nebular lines due to high escape of ionising photons. The small Lyα
peak separation of 220 ± 20 km s−1 implies a low HI column density and an ionising
photon escape fraction of ≈15 − 30%, providing the first direct evidence that
such galaxies contribute actively to the reionisation of the Universe at z >  6.
Based on simple estimates, we find that COLA1 could have provided just enough
photons to reionise its own ≈0.3 pMpc (2.3 cMpc) bubble, allowing the blue Lyα
line to be observed. However, we also discuss alternative scenarios explaining
the detected double peaked nature of COLA1. Our results show that future high-resolution
observations of statistical samples of double peaked LAEs at z >  5 are a promising
probe of the occurrence of ionised regions around galaxies in the EoR.

text: We investigate the morphology of the [C II] emission in a sample of ‘normal’
    star-forming galaxies at 5 < z < 7.2 in relation to their UV (rest-frame) counterpart.
We use new Atacama Large Millimetre/submillimetre Array (ALMA) observations of
galaxies at z ∼ 6–7, as well as a careful re-analysis of archival ALMA data. In
total 29 galaxies were analysed, 21 of which are detected in [C II]. For several
of the latter the [C II] emission breaks into multiple components. Only a fraction
of these [C II] components, if any, is associated with the primary UV systems,
while the bulk of the [C II] emission is associated either with fainter UV components,
or not associated with any UV counterpart at the current limits. By taking into
account the presence of all these components, we find that the L[CII]–SFR (star
formation rate) relation at early epochs is fully consistent with the local relation,
but it has a dispersion of 0.48 ± 0.07 dex, which is about two times larger than
observed locally. We also find that the deviation from the local L[CII]–SFR relation
has a weak anticorrelation with the EW(Ly α). The morphological analysis also
reveals that [C II] emission is generally much more extended than the UV emission.
As a consequence, these primordial galaxies are characterized by a [C II] surface
brightness generally much lower than expected from the local Σ[CII]−ΣSFR relation.
These properties are likely a consequence of a combination of different effects,
namely gas metallicity, [C II] emission from obscured star-forming regions, strong
variations of the ionization parameter, and circumgalactic gas in accretion or
    ejected by these primeval galaxies.

text: We present and explore deep narrow- and medium-band data obtained with the
Subaru and the Isaac Newton Telescopes in the ∼2 deg2 COSMOS field. We use these
data as an extremely wide, low-resolution (R ∼ 20–80) Integral Field Unit survey
to slice through the COSMOS field and obtain a large sample of ∼4000 Ly α emitters
(LAEs) from z ∼ 2 to 6 in 16 redshift slices (SC4K). We present new Ly α luminosity
functions (LFs) covering a comoving volume of ∼108 Mpc3. SC4K extensively complements
ultradeep surveys, jointly covering over 4 dex in Ly α luminosity and revealing
    a global (2.5 < z < 6) synergy LF with α=−1.93+0.12−0.12, log10Φ∗Lyα=−3.45+0.22−0.29 Mpc−3,
and log10L∗Lyα=42.93+0.15−0.11 erg s−1. The Schechter component of the Ly α LF
    reveals a factor ∼5 rise in L∗Lyα and a ∼7 × decline in Φ∗Lyα from z ∼ 2 to 6.
The data reveal an extra power-law (or Schechter) component above LLy α ≈ 1043.3 erg s−1
at z ∼ 2.2–3.5 and we show that it is partially driven by X-ray and radio active
galactic nucleus (AGN), as their Ly α LF resembles the excess. The power-law component
vanishes and/or is below our detection limits above z > 3.5, likely linked with
the evolution of the AGN population. The Ly α luminosity density rises by a factor
    ∼2 from z ∼ 2 to 3 but is then found to be roughly constant (1.1+0.2−0.2×1040 erg s−1 Mpc−3)
to z ∼ 6, despite the ∼0.7 dex drop in ultraviolet (UV) luminosity density. The
Ly α/UV luminosity density ratio rises from 4 ± 1 per cent to 30 ± 6 per cent
from z ∼ 2.2 to 6. Our results imply a rise of a factor of ≈2 in the global ionization
    efficiency (ξion) and a factor ≈4 ± 1 in the Ly α escape fraction from z ∼ 2 to
6, hinting for evolution in both the typical burstiness/stellar populations and
even more so in the typical interstellar medium conditions allowing Ly α photons
    to escape.

text: Glaciers in the high mountains of Asia provide an important water resource
for millions of people. Many of these glaciers are partially covered by rocky
debris, which protects the ice from solar radiation and warm air. However, studies
have found that the surface of these debris-covered glaciers is actually lowering
as fast as glaciers without debris. Water ponded on the surface of the glaciers
may be partially responsible, as water can absorb atmospheric energy very efficiently.
However, the overall effect of these ponds has not been thoroughly assessed yet.
We study a valley in Nepal for which we have extensive weather measurements, and
we use a numerical model to calculate the energy absorbed by ponds on the surface
of the glaciers over 6 months. As we have not observed each individual pond thoroughly,
we run the model 5,000 times with different setups. We find that ponds are extremely
important for glacier melt and absorb energy 14 times as quickly as the debris-covered
ice. Although the ponds account for 1% of the glacier area covered by rocks, and
only 0.3% of the total glacier area, they absorb enough energy to account for
one eighth of the whole valley's ice loss.

text: Recent advances in the fabrication of nanostructures and nanoscale features
in metasurfaces offer new prospects for generating visible light emission from
low-energy electrons. Here we present the experimental observation of visible
light emission from low-energy free electrons interacting with nanoscale periodic
surfaces through the Smith–Purcell (SP) effect. We demonstrate SP light emission
from nanoscale gratings with periodicity as small as 50 nm, enabling the observation
of tunable visible radiation from low-energy electrons (1.5 to 6 keV), an order
of magnitude lower in energy than previously reported. We study the emission wavelength
and intensity dependence on the grating pitch and electron energy, showing agreement
between experiment and theory. Our results open the way to the production of SP-based
nanophotonics integrated devices. Built inside electron microscopes, SP sources
could enable the development of novel electron–optical correlated spectroscopic
techniques and facilitate the observation of new quantum effects in light sources.

text: Confining molecules to volumes only slightly larger than the molecules themselves
can profoundly alter their properties. Molecular switches—entities that can be
toggled between two or more forms upon exposure to an external stimulus—often
require conformational freedom to isomerize. Therefore, placing these switches
in confined spaces can render them non-operational. To preserve the switchability
of these species under confinement, we work with a water-soluble coordination
cage that is flexible enough to adapt its shape to the conformation of the encapsulated
guest. We show that owing to its flexibility, the cage is not only capable of
    accommodating—and solubilizing in water—several light-responsive spiropyran-based
molecular switches, but, more importantly, it also provides an environment suitable
for the efficient, reversible photoisomerization of the bound guests. Our findings
pave the way towards studying various molecular switching processes in confined
    environments.

text: Stars stripped of their hydrogen-rich envelope through interaction with a
binary companion are generally not considered when accounting for ionizing radiation
from stellar populations, despite the expectation that stripped stars emit hard
ionizing radiation, form frequently, and live 10–100 times longer than single
massive stars. We compute the first grid of evolutionary and spectral models specially
made for stars stripped in binaries for a range of progenitor masses (2–20 M⊙)
and metallicities ranging from solar to values representative for pop II stars.
For stripped stars with masses in the range 0.3–7 M⊙, we find consistently high
    effective temperatures (20 000–100 000 K, increasing with mass), small radii (0.2–1
R⊙), and high bolometric luminosities, comparable to that of their progenitor
before stripping. The spectra show a continuous sequence that naturally bridges
subdwarf-type stars at the low-mass end and Wolf-Rayet-like spectra at the high-mass
end. For intermediate masses we find hybrid spectral classes showing a mixture
of absorption and emission lines. These appear for stars with mass-loss rates
of 10−8−10−6 M⊙ yr−1, which have semi-transparent atmospheres. At low metallicity,
substantial hydrogen-rich layers are left at the surface and we predict spectra
that resemble O-type stars instead. We obtain spectra undistinguishable from subdwarfs
for stripped stars with masses up to 1.7 M⊙, which questions whether the widely
adopted canonical value of 0.47 M⊙ is uniformly valid. Only a handful of stripped
stars of intermediate mass have currently been identified observationally. Increasing
this sample will provide necessary tests for the physics of interaction, internal
mixing, and stellar winds. We use our model spectra to investigate the feasibility
to detect stripped stars next to an optically bright companion and recommend systematic
searches for their UV excess and possible emission lines, most notably HeII λ4686
in the optical and HeII λ1640 in the UV. Our models are publicly available for
further investigations or inclusion in spectral synthesis simulations.

text: "Molecular chirality plays an essential role in most biochemical processes.
The observation and quantification of chirality-sensitive signals, however, remains
extremely challenging, especially on ultrafast timescales and in dilute media.
Here, we describe the experimental realization of an all-optical and ultrafast
scheme for detecting chiral dynamics in molecules. This technique is based on
high-harmonic generation by a combination of two-color counterrotating femtosecond
laser pulses with polarization states tunable from linear to circular. We demonstrate
two different implementations of chiral-sensitive high-harmonic spectroscopy on
an ensemble of randomly oriented methyloxirane molecules in the gas phase. Using
two elliptically polarized fields, we observe that the ellipticities maximizing
the harmonic signal reach up to \r\n4.4\r\n±\r\n0.2\r\n%\r\n (at 17.6 eV). Using
two circularly polarized fields, we observe circular dichroisms ranging up to
\r\n13\r\n±\r\n6\r\n%\r\n (28.3–33.1 eV). Our theoretical analysis confirms that
the observed chiral response originates from subfemtosecond electron dynamics
driven by the magnetic component of the driving laser field. This assignment is
supported by the experimental observation of a strong intensity dependence of
the chiral effects and its agreement with theory. We moreover report and explain
a pronounced variation of the signal strength and dichroism with the driving-field
ellipticities and harmonic orders. Finally, we demonstrate the sensitivity of
the experimental observables to the shape of the electron hole. This technique
for chiral discrimination will yield femtosecond temporal resolution when integrated
in a pump-probe scheme and subfemtosecond resolution on chiral charge migration
    in a self-probing scheme."

text: The task of a monitor is to watch, at run-time, the execution of a reactive
system, and signal the occurrence of a safety violation in the observed sequence
of events. While finite-state monitors have been studied extensively, in practice,
monitoring software also makes use of unbounded memory. We define a model of automata
equipped with integer-valued registers which can execute only a bounded number
of instructions between consecutive events, and thus can form the theoretical
basis for the study of infinite-state monitors. We classify these register monitors
according to the number k of available registers, and the type of register instructions.
In stark contrast to the theory of computability for register machines, we prove
    that for every k 1, monitors with k + 1 counters (with instruction set 〈+1, =〉)
are strictly more expressive than monitors with k counters. We also show that
adder monitors (with instruction set 〈1, +, =〉) are strictly more expressive than
counter monitors, but are complete for monitoring all computable safety -languages
for k = 6. Real-time monitors are further required to signal the occurrence of
a safety violation as soon as it occurs. The expressiveness hierarchy for counter
monitors carries over to real-time monitors. We then show that 2 adders cannot
simulate 3 counters in real-time. Finally, we show that real-time adder monitors
with inequalities are as expressive as real-time Turing machines.

text: Starting from any given rational-sided, right triangle, for example, the (3,4,5)-triangle
with area 6, we use Euclidean geometry to show that there are infinitely many
other rational-sided, right triangles of the same area. We show further that the
set of all such triangles of a given area is finitely generated under our geometric
construction. Such areas are known as “congruent numbers” and have a rich history
in which all the results in this article have been proved and far more. Yet, as
far as we can tell, this seems to be the first exploration using this kind of
    geometric technique.

text: "We consider the problem of expected cost analysis over nondeterministic probabilistic
programs, which aims at automated methods for analyzing the resource-usage of
such programs. Previous approaches for this problem could only handle nonnegative
bounded costs. However, in many scenarios, such as queuing networks or analysis
of cryptocurrency protocols, both positive and negative costs are necessary and
the costs are unbounded as well.\r\n\r\nIn this work, we present a sound and efficient
approach to obtain polynomial bounds on the expected accumulated cost of nondeterministic
probabilistic programs. Our approach can handle (a) general positive and negative
costs with bounded updates in variables; and (b) nonnegative costs with general
updates to variables. We show that several natural examples which could not be
handled by previous approaches are captured in our framework.\r\n\r\nMoreover,
our approach leads to an efficient polynomial-time algorithm, while no previous
approach for cost analysis of probabilistic programs could guarantee polynomial
runtime. Finally, we show the effectiveness of our approach by presenting experimental
results on a variety of programs, motivated by real-world applications, for which
we efficiently synthesize tight resource-usage bounds."

text: Lesion and electrode location verification are traditionally done via histological
examination of stained brain slices, a time-consuming procedure that requires
manual estimation. Here, we describe a simple, straightforward method for quantifying
lesions and locating electrodes in the brain that is less laborious and yields
more detailed results. Whole brains are stained with osmium tetroxide, embedded
in resin, and imaged with a micro-CT scanner. The scans result in 3D digital volumes
of the brains with resolutions and virtual section thicknesses dependent on the
sample size (12-15 and 5-6 µm per voxel for rat and zebra finch brains, respectively).
Surface and deep lesions can be characterized, and single tetrodes, tetrode arrays,
electrolytic lesions, and silicon probes can also be localized. Free and proprietary
software allows experimenters to examine the sample volume from any plane and
segment the volume manually or automatically. Because this method generates whole
brain volume, lesions and electrodes can be quantified to a much higher degree
than in current methods, which will help standardize comparisons within and across
    studies.
