In the present study, the possible changes in the expression of metabotropic glutamate receptors (mGluRs) were examined in the basal ganglia of a primate model for Parkinson's disease. When the patterns of immunohistochemical localization of mGluRs in monkeys administered systemically with 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) were analysed in comparison with normal controls, we found that expression of mGluR1α, but not of other subtypes, was significantly reduced in the internal and external segments of the globus pallidus and the substantia nigra pars reticulata. To elucidate the functional role of mGluR1 in the control of pallidal neuron activity, extracellular unit recordings combined with intrapallidal microinjections of mGluR1-related agents were then performed in normal and parkinsonian monkeys. In normal awake conditions, the spontaneous firing rates of neurons in the pallidal complex were increased by DHPG, a selective agonist of group I mGluRs, whereas they were decreased by AIDA, a selective antagonist of group I mGluRs, or LY367385, a selective antagonist of mGluR1. These electrophysiological data strongly indicate that the excitatory mechanism of pallidal neurons by glutamate is mediated at least partly through mGluR1. The effects of the mGluR1-related agents on neuronal firing in the internal pallidal segment became rather obscure after MPTP treatment. Our results suggest that the specific down-regulation of pallidal and nigral mGluR1 ot in the parkinsonian state may exert a compensatory action to reverse the overactivity of the subthalamic nucleus-derived glutamatergic input that is generated in the disease."}],"type":"journal_article","oa_version":"None","article_type":"original","OA_type":"closed access","language":[{"iso":"eng"}],"publist_id":"4240","issue":"12","publication":"European Journal of Neuroscience","publication_identifier":{"eissn":["1460-9568"],"issn":["0953-816X"]}},{"date_updated":"2026-08-06T13:45:05Z","user_id":"317138e5-6ab7-11ef-aa6d-ffef3953e345","page":"3034 - 3042","citation":{"apa":"Nyíri, G., Szabadits, E., Cserép, C., Mackie, K., Shigemoto, R., & Freund, T. The range of receptors for endogenous modulators expressed on these neurons is unclear. Here we describe GABAB 1a/b receptor (GABABR) and type 1 cannabinoid receptor (CB1R) expression in rat septal cholinergic [i.e. choline acetyltransferase (ChAT)-positive] cells. Using double immunofluorescent staining, we found that almost two-thirds of the cholinergic cells in the rat medial septum were GABABR positive, and that these cells had significantly larger somata than did GABABR-negative cholinergic neurons. We detected CB1R labelling in somata after axonal protein transport was blocked by colchicine. In these animals about one-third of the cholinergic cells were CB1R positive. These cells again had larger somata than CB1R-negative cholinergic neurons. The analyses confirmed that the size of GABABR-positive and CB 1R-positive cholinergic cells were alike, and all CB 1R-positive cholinergic cells were GABABR positive as well. CB1R-positive cells were invariably ChAT positive. All retrogradely labelled septohippocampal cholinergic cells were positive for GABABR and at least half of them also for CB1R. These data shed light on the existence of at least two cholinergic cell types in the medial septum: one expresses GABABR and CB1R, has large somata and projects to the hippocampus, whereas the other is negative for GABABR and CB1R and has smaller somata. The results also suggest that cholinergic transmission in the hippocampus is fine-tuned by endocannabinoid signalling."}],"title":"GABAB and CB1 cannabinoid receptor expression identifies two types of septal cholinergic neurons","_id":"2650","author":[{"full_name":"Nyíri, Gábor","last_name":"Nyíri","first_name":"Gábor"},{"full_name":"Szabadits, Eszter","last_name":"Szabadits","first_name":"Eszter"},{"last_name":"Cserép","first_name":"Csaba","full_name":"Cserép, Csaba"},{"full_name":"Mackie, Ken","first_name":"Ken","last_name":"Mackie"},{"id":"499F3ABC-F248-11E8-B48F-1D18A9856A87","first_name":"Ryuichi","last_name":"Shigemoto","orcid":"0000-0001-8761-9444","full_name":"Shigemoto, Ryuichi"},{"full_name":"Freund, Tamás","first_name":"Tamás","last_name":"Freund"}],"date_created":"2018-12-11T11:58:52Z","status":"public","language":[{"iso":"eng"}],"publist_id":"4247","OA_type":"closed access","publication_identifier":{"eissn":["1460-9568"],"issn":["0953-816X"]},"publication":"European Journal of Neuroscience","issue":"11","fulldoi":"https://doi.org/10.1111/j.1460-9568.2005.04146.x","date_published":"2005-06-01T00:00:00Z","article_processing_charge":"No","publication_status":"published","pmid":1,"year":"2005","intvolume":" 21","volume":21,"doi":"10.1111/j.1460-9568.2005.04146.x","external_id":{"pmid":["15978014 "]},"publisher":"Wiley-Blackwell","month":"06","day":"01"},{"publist_id":"4248","language":[{"iso":"eng"}],"OA_type":"closed access","publication_identifier":{"eissn":["1460-9568"],"issn":["0953-816X"]},"publication":"European Journal of Neuroscience","issue":"8","user_id":"317138e5-6ab7-11ef-aa6d-ffef3953e345","date_updated":"2026-08-06T13:48:42Z","page":"2073 - 2082","citation":{"chicago":"Luján, Rafael, José Albasanz, Ryuichi Shigemoto, and José Juíz. “Preferential Localization of the Hyperpolarization-Activated Cyclic Nucleotide-Gated Cation Channel Subunit HCN1 in Basket Cell Terminals of the Rat Cerebellum.” European Journal of Neuroscience. In this study, we used immunoblotting and immunohistochemical techniques to reveal the developmental expression and subcellular distribution of the HCN1 subunit in the cerebellar cortex. During postnatal development, the spatio-temporal expression of HCN1 correlated well with the morphological events occurring during the ontogenesis of cerebellar interneurons. Using immunoblotting techniques, HCN1 was weakly detected during the first postnatal week and continued to increase throughout postnatal development, peaking at postnatal day (P)15. At the light-microscopic level, HCN1 immunoreactivity was very weak until P7 whereas from P10-12 to adulthood it was strongly detected in the lower third of the molecular layer and in the Purkinje cell layer. HCN1 was present in axons running through the molecular layer and in the pericellular basket around Purkinje cells at P12, but in the periaxonal plexus (the pinceau) surrounding their initial segment only after P15. Using immunofluorescence, HCN1 colocalized with GAD65 and synaptophysin, demonstrating that the subunit was present in inhibitory axons and axon terminals. At the electron-microscopic level, in adulthood, HCN1 immunoparticles were detected at postsynaptic sites in basket and Purkinje cells but most immunoparticles were found at presynaptic sites in basket cell axons and in terminals. In the axon terminals, the distribution of HCN1 was relatively uniform along the extrasynaptic plasma membrane; this was confirmed using quantitative techniques. The present findings suggest that HCN1 channels may provide a significant route for modulating co-ordinated cerebellar synaptic transmission through basket cells.","lang":"eng"}],"extern":"1","_id":"2648","title":"Preferential localization of the hyperpolarization-activated cyclic nucleotide-gated cation channel subunit HCN1 in basket cell terminals of the rat cerebellum","status":"public","date_created":"2018-12-11T11:58:52Z","author":[{"full_name":"Luján, Rafael","first_name":"Rafael","last_name":"Luján"},{"full_name":"Albasanz, José","first_name":"José","last_name":"Albasanz"},{"last_name":"Shigemoto","first_name":"Ryuichi","id":"499F3ABC-F248-11E8-B48F-1D18A9856A87","orcid":"0000-0001-8761-9444","full_name":"Shigemoto, Ryuichi"},{"first_name":"José","last_name":"Juíz","full_name":"Juíz, José"}],"volume":21,"doi":"10.1111/j.1460-9568.2005.04043.x","external_id":{"pmid":["15869503 "]},"month":"04","publisher":"Wiley-Blackwell","day":"01","fulldoi":"https://doi.org/10.1111/j.1460-9568.2005.04043.x","date_published":"2005-04-01T00:00:00Z","article_processing_charge":"No","pmid":1,"year":"2005","publication_status":"published","intvolume":" 21"},{"article_type":"original","type":"journal_article","oa_version":"None","extern":"1","abstract":[{"lang":"eng","text":"The release of GABA in synapses is modulated by presynaptic metabotropic glutamate receptors (mGluRs). We tested whether GABA release to identified hippocampal neurons is influenced by group III mGluR activation using the agonist L-(+)-2-amino-4-phosphonobutyric acid (L-AP4) on inhibitory postsynaptic currents (IPSCs) evoked in CA1 interneurons and pyramidal cells. In interneurons, characterized with biocytin and immunolabelling for somatostatin, evoked IPSCs were depressed by 50 μM L-AP4 (activating mGluR4 and 8) to 68±6% of control, but they were rarely depressed in pyramidal cells (96±4% of control). At 300-500 μM concentration (activating mGluR4, 7 and 8), L-AP4 depressed IPSCs in both interneurons (to 70±6%) and pyramidal cells (to 67±4%). The change in trial-to-trial variability and in paired-pulse depression indicated a presynaptic action. In interneurons, the degree of IPSC depression was variable (to 9-87%), and a third of IPSCs were not affected by L-AP4. The L-AP4-evoked IPSC depression was blocked by LY341495. The depression of IPSCs was similar in O-LM cells and other interneurons. The lack of cell-type selectivity and the similar efficacy of different concentrations of L-AP4 suggest that several group III mGluRs are involved in the depression of IPSCs. Electron microscopic immunocytochemistry confirmed that mGluR4, mGluR7a and mGluR8a occur in the presynaptic active zone of GABAergic terminals on interneurons, but not on those innervating pyramidal cells. The high variability of L-AP4-evoked IPSC suppression is in line with the selective expression of presynaptic mGluRs by several distinct types of GABAergic neuron innervating each interneuron type."}],"_id":"2644","title":"Depression of GABAergic input to identified hippocampal neurons by group III metabotropic glutamate receptors in the rat","author":[{"last_name":"Kogo","first_name":"Naoki","full_name":"Kogo, Naoki"},{"first_name":"Yannis","last_name":"Dalezios","full_name":"Dalezios, Yannis"},{"first_name":"Marco","last_name":"Capogna","full_name":"Capogna, Marco"},{"full_name":"Ferraguti, Francesco","first_name":"Francesco","last_name":"Ferraguti"},{"orcid":"0000-0001-8761-9444","full_name":"Shigemoto, Ryuichi","last_name":"Shigemoto","first_name":"Ryuichi","id":"499F3ABC-F248-11E8-B48F-1D18A9856A87"},{"full_name":"Somogyi, Péter","first_name":"Péter","last_name":"Somogyi"}],"date_created":"2018-12-11T11:58:50Z","status":"public","user_id":"317138e5-6ab7-11ef-aa6d-ffef3953e345","date_updated":"2026-09-22T13:19:16Z","page":"2727 - 2740","citation":{"short":"N. All VGLUT3-positive somata were immunoreactive for CCK, and very rarely, also for calbindin; none was positive for parvalbumin, calretinin, VIP or somatostatin. In the CA1 area, 26.8 ± 0.7% of CCK-positive interneuron somata were VGLUT3-positive, a nonoverlapping 22.8 ± 1.9% were calbindin-positive, 10.7 ± 2.5% VIP-positive and the rest were only CCK-positive. The patterns of coexpression were similar in the CA3 area, the dentate gyrus and the isocortex. Immunoreactivity for VGLUT3 was undetectable in pyramidal and dentate granule cells. Boutons colabelled for VGLUT3, CCK and GAD were most abundant in the cellular layers of the hippocampus and in layers II-III of the isocortex. Large VGLUT3-labelled boutons at the border of strata radiatum and lacunosum-moleculare in the CA1 area were negative for GAD, but were labelled for vesicular monoamine transporter type 2, plasmalemmal serotonin transporter or serotonin. No colocalization was found in terminals between VGLUT3 and parvalbumin, vesicular acetylcholine transporter and group III (mGluR7a,b; mGluR8a,b) metabotropic glutamate receptors. In stratum radiatum and the isocortex, VGLUT3-positive but GAD-negative boutons heavily innervated the soma and proximal dendrites of some VGLUT3- or calbindin-positive interneurons. The results suggest that boutons coexpressing VGLUT3, CCK and GAD originate from CCK-positive basket cells, which are VIP-immunonegative. Other VGLUT3-positive boutons immunopositive for serotonergic markers but negative for GAD probably originate from the median raphe nucleus and innervate select interneurons. The presumed amino acid substrate of VGLUT3 may act on presynaptic kainate or group II metabotropic glutamate receptors."}],"extern":"1","article_type":"original","type":"journal_article","oa_version":"None","status":"public","date_created":"2018-12-11T11:58:49Z","author":[{"full_name":"Somogyi, Jozsef","last_name":"Somogyi","first_name":"Jozsef"},{"first_name":"Agnès","last_name":"Baude","full_name":"Baude, Agnès"},{"full_name":"Omori, Yuko","last_name":"Omori","first_name":"Yuko"},{"first_name":"Hidemi","last_name":"Shimizu","full_name":"Shimizu, Hidemi"},{"full_name":"El Mestikawy, Salah","last_name":"El Mestikawy","first_name":"Salah"},{"last_name":"Fukaya","first_name":"Masahiro","full_name":"Fukaya, Masahiro"},{"last_name":"Shigemoto","first_name":"Ryuichi","id":"499F3ABC-F248-11E8-B48F-1D18A9856A87","orcid":"0000-0001-8761-9444","full_name":"Shigemoto, Ryuichi"},{"first_name":"Masahiko","last_name":"Watanabe","full_name":"Watanabe, Masahiko"},{"first_name":"Péter","last_name":"Somogyi","full_name":"Somogyi, Péter"}],"_id":"2639","title":"GABAergic basket cells expressing cholecystokinin contain vesicular glutamate transporter type 3 (VGLUT3) in their synaptic terminals in hippocampus and isocortex of the rat","language":[{"iso":"eng"}],"publist_id":"4260","OA_type":"closed access","publication_identifier":{"eissn":["1460-9568"],"issn":["0953-816X"]},"issue":"3","publication":"European Journal of Neuroscience"},{"publication_identifier":{"eissn":["1460-9568"],"issn":["0953-816X"]},"issue":"8","publication":"European Journal of Neuroscience","publist_id":"4259","language":[{"iso":"eng"}],"OA_type":"closed access","abstract":[{"text":"Among various types of low- and high-threshold calcium channels, the high voltage-activated P/Q-type channel is the most abundant in the cerebellum. These P/Q-type channels are involved in the regulation of neurotransmitter release and in the integration of dendritic inputs. We used an antibody specific for the α1A subunit of the P/Q-type channel in quantitative pre-embedding immunogold labelling combined with three-dimensional reconstruction to reveal the subcellular distribution of pre- and postsynaptic P/Q-type channels in the rat cerebellum. At the light microscopic level, immunoreactivity for the α1A protein was prevalent in the molecular layer, whereas immunostaining was moderate in the somata of Purkinje cells and weak in the granule cell layer. At the electron microscopic level, the most intense Immunoreactivity for the α1A subunit was found in the presynaptic active zone of parallel fibre varicosities. The dendritic spines of Purkinje cells were also strongly labelled with the highest density of immunoparticles detected within 180 nm from the edge of the asymmetrical parallel fibre-Purkinje cell synapses. By contrast, the immunolabelling was sparse in climbing fibre varicosities and axon terminals of GABAergic cells, and weak and diffuse in dendritic shafts of Purkinje cells. The association of the α1A subunit with the glutamatergic parallel fibre-Purkinje cell synapses suggests that presynaptic channels have a major role in the mediation of excitatory neurotransmission, whereas postsynaptic channels are likely to be involved in depolarization-induced generation of local calcium transients in Purkinje cells.","lang":"eng"}],"extern":"1","oa_version":"None","type":"journal_article","article_type":"original","date_created":"2018-12-11T11:58:48Z","author":[{"first_name":"Ákos","last_name":"Kulik","full_name":"Kulik, Ákos"},{"full_name":"Nakadate, Kazuhiko","last_name":"Nakadate","first_name":"Kazuhiko"},{"full_name":"Hagiwara, Akari","first_name":"Akari","last_name":"Hagiwara"},{"full_name":"Fukazawa, Yugo","last_name":"Fukazawa","first_name":"Yugo"},{"last_name":"Luján","first_name":"Rafael","full_name":"Luján, Rafael"},{"last_name":"Saito","first_name":"Hiromitsu","full_name":"Saito, Hiromitsu"},{"first_name":"Noboru","last_name":"Suzuki","full_name":"Suzuki, Noboru"},{"full_name":"Futatsugi, Akira","last_name":"Futatsugi","first_name":"Akira"},{"full_name":"Mikoshiba, Katsuhiko","first_name":"Katsuhiko","last_name":"Mikoshiba"},{"last_name":"Frotscher","first_name":"Michael","full_name":"Frotscher, Michael"},{"full_name":"Shigemoto, Ryuichi","orcid":"0000-0001-8761-9444","id":"499F3ABC-F248-11E8-B48F-1D18A9856A87","last_name":"Shigemoto","first_name":"Ryuichi"}],"status":"public","title":"Immunocytochemical localization of the α1A subunit of the P/Q-type calcium channel in the rat cerebellum","_id":"2638","date_updated":"2026-09-30T12:53:54Z","user_id":"317138e5-6ab7-11ef-aa6d-ffef3953e345","page":"2169 - 2178","citation":{"ama":"Kulik Á, Nakadate K, Hagiwara A, et al. We have studied activity-dependent changes in mGluR1 function in mouse cultured Purkinje neurons. Depolarizing stimulation potentiated Ca2+ and current responses to an mGluR1 agonist for several hours in the cultured Purkinje neurons. It also blocked internalization of mGluR1 and increased the number of mGluR1s on the cell membrane. We found that depolarization simultaneously increased transcription of Homer1a in Purkinje neurons. Homer1a inhibited internalization and increased cell-surface expression of mGluR1 when coexpressed in human embryonic kidney (HEK)-293 cells. Depolarization-induced Homer1a expression in Purkinje neurons was blocked by a mitogen-activated protein kinase (MAPK) inhibitor. Changes in internalization and mGluR1-mediated Ca2+ response were also blocked by inhibition of MAPK activity, suggesting that localization and activity of mGluR1 were regulated in the same signalling pathway as Homer1a expression. It is thus suggested that depolarization of the Purkinje neuron leads to the increment in mGluR1 responsiveness through MAPK activity and induction of Homer1a expression, which increases active mGluR1 on the cell surface by blocking internalization of mGluR1."}],"OA_type":"closed access","language":[{"iso":"eng"}],"publist_id":"4273","publication":"European Journal of Neuroscience","issue":"5","scopus_import":"1","publication_identifier":{"issn":["0953-816X"],"eissn":["1460-9568"]}},{"day":"23","publisher":"Wiley","month":"06","external_id":{"pmid":["12823458"]},"doi":"10.1046/j.1460-9568.2003.02697.x","volume":17,"intvolume":" 17","publication_status":"published","quality_controlled":"1","pmid":1,"year":"2003","article_processing_charge":"No","date_published":"2003-06-23T00:00:00Z","fulldoi":"https://doi.org/10.1046/j.1460-9568.2003.02697.x","scopus_import":"1","issue":"12","publication":"European Journal of Neuroscience","publication_identifier":{"issn":["0953-816X"],"eissn":["1460-9568"]},"OA_type":"closed access","publist_id":"4269","language":[{"iso":"eng"}],"author":[{"last_name":"Somogyi","first_name":"Péter","full_name":"Somogyi, Péter"},{"full_name":"Dalezios, Yannis","first_name":"Yannis","last_name":"Dalezios"},{"last_name":"Luján","first_name":"Rafael","full_name":"Luján, Rafael"},{"first_name":"John","last_name":"Roberts","full_name":"Roberts, John"},{"full_name":"Watanabe, Masahiko","last_name":"Watanabe","first_name":"Masahiko"},{"id":"499F3ABC-F248-11E8-B48F-1D18A9856A87","first_name":"Ryuichi","last_name":"Shigemoto","full_name":"Shigemoto, Ryuichi","orcid":"0000-0001-8761-9444"}],"status":"public","date_created":"2018-12-11T11:58:46Z","title":"High level of mGluR7 in the presynaptic active zones of select populations of GABAergic terminals innervating interneurons in the rat hippocampus","_id":"2629","abstract":[{"text":"The release of neurotransmitters is modulated by presynaptic metabotropic glutamate receptors (mGluRs), which show a highly selective expression and subcellular location in glutamatergic terminals in the hippocampus. Using immunocytochemistry, we investigated whether one of the receptors, mGluR7, whose level of expression is governed by the postsynaptic target, was present in GABAergic terminals and whether such terminals targeted particular cells. A total of 165 interneuron dendritic profiles receiving 466 synapses (82% mGluR7a-positive) were analysed. The presynaptic active zones of most GAD-(77%) or GABA-positive (94%) synaptic boutons on interneurons innervated by mGluR7a-enriched glutamatergic terminals (mGluR7a-decorated) were immunopositive for mGluR7a. GABAergic terminals on pyramidal cells and most other interneurons in str. oriens were mGluR7a-immunonegative. The mGluR7a-decorated cells were mostly somatostatin- and mGluR1α-immunopositive neurons in str. oriens and the alveus. Their GABAergic input mainly originated from VIP-positive terminals, 90% of which expressed high levels of mGluR7a in the presynaptic active zone. Parvalbumin-positive synaptic terminals were rare on mGluR7a-decorated cells, but on these neurons 73% of them were mGluR7a-immunopositive. Some type II synapses innervating interneurons were immunopositive for mGluR7b, as were some type I synapses. Because not all target cells of VIP-positive neurons are known it has not been possible to determine whether mGluR7 is expressed in a target-cell-specific manner in the terminals of single GABAergic cells. The activation of mGluR7 may decrease GABA release to mGluR7-decorated cells at times of high pyramidal cell activity, which elevates extracellular glutamate levels. Alternatively, the presynaptic receptor may be activated by as yet unidentified endogenous ligands released by the GABAergic terminals or the postsynaptic dendrites.","lang":"eng"}],"extern":"1","oa_version":"None","type":"journal_article","article_type":"original","citation":{"chicago":"Somogyi, Péter, Yannis Dalezios, Rafael Luján, John Roberts, Masahiko Watanabe, and Ryuichi Shigemoto. “High Level of MGluR7 in the Presynaptic Active Zones of Select Populations of GABAergic Terminals Innervating Interneurons in the Rat Hippocampus.” European Journal of Neuroscience. Knowledge of the cellular and subcellular distributions of hyperpolarization-activated and cyclic nucleotide-gated channel (HCN) subunits is necessary for understanding the role of Ih in olfactory network activities. Using light microscopic immunocytochemistry, we demonstrate strong HCN1 labelling of the glomerular layer and moderate staining of granule cell, internal and external plexiform layers of the rat main olfactory bulb. In the glomerular layer, among many unlabelled neurons, two distinct subpopulations of juxtaglomerular cells are labelled. Approximately 10% of the juxtaglomerular cells strongly express HCN1. These small diameter cells are immunoreactive for GABA and comprise a subpopulation of periglomerular cells. An additional subset of juxtaglomerular cells (≈ 1%) expresses low levels of HCN1. They are large in diameter, GABA immunonegative but immunopositive for vesicular glutamate transporter 2, characterizing them as external tufted cells. Quantitative immunogold localization revealed that the somatic plasma membranes of periglomerular cells contain approximately four times more HCN1 labelling than those of external tufted cells. Unlike in cortical pyramidal cells, immunogold density for HCN1 does not significantly differ in somatic and dendritic plasma membranes of external tufted cells, indicating that post-synaptic potentials arriving at proximal and distal dendrites are modulated by the same density of I h. Our results demonstrate a cell type-dependent expression of HCN1 in the olfactory bulb and predict a differential contribution of distinct juxtaglomerular cell types to network oscillations."}],"extern":"1","oa_version":"None","type":"journal_article","article_type":"original","author":[{"last_name":"Holderith","first_name":"Noémi","full_name":"Holderith, Noémi"},{"first_name":"Ryuichi","last_name":"Shigemoto","id":"499F3ABC-F248-11E8-B48F-1D18A9856A87","full_name":"Shigemoto, Ryuichi","orcid":"0000-0001-8761-9444"},{"first_name":"Zoltán","last_name":"Nusser","full_name":"Nusser, Zoltán"}],"date_created":"2018-12-11T11:58:47Z","status":"public","title":"Cell type-dependent expression of HCN1 in the main olfactory bulb","_id":"2632","language":[{"iso":"eng"}],"publist_id":"4266","OA_type":"closed access","publication_identifier":{"eissn":["1460-9568"],"issn":["0953-816X"]},"scopus_import":"1","issue":"2","publication":"European Journal of Neuroscience","fulldoi":"https://doi.org/10.1046/j.1460-9568.2003.02756.x","article_processing_charge":"No","date_published":"2003-07-01T00:00:00Z","intvolume":" 18","publication_status":"published","quality_controlled":"1","year":"2003","pmid":1,"doi":"10.1046/j.1460-9568.2003.02756.x","volume":18,"publisher":"Wiley-Blackwell","month":"07","external_id":{"pmid":["12887416"]},"day":"01"},{"language":[{"iso":"eng"}],"publist_id":"4276","publication":"European Journal of Neuroscience","scopus_import":"1","issue":"11","acknowledgement":"The authors are grateful to Dr Marco Sassoe-Pogneto for his comments on a previous version of the manuscript. At the light microscopic level, both GABABR1 and GABABR2 were detected as early as E14. During prenatal development, both subtypes were expressed highly in the cortical plate. Using double immunofluorescence, GABABR1 colocalized with GABABR2 in neurons of the marginal zone and subplate, indicating that these proteins are coexpressed and could be forming functional GABABRs during prenatal development in vivo. In contrast, only GABABR1 but not GABABR2 was detected in the tangentially migratory cells in the lower intermediate zone. During postnatal development, immunoreactivity for GABABR1 and GABABR2 was distributed mainly in pyramidal cells. Discrete GABABR1-immunopositive cell bodies of interneurons were present throughout the neocortex. In addition, GABABR1 but not GABABR2 was found in identified Cajal-Retzius cells in layer I. At the electron microscopic level, immunoreactivity for GABABR1 and GABABR2 was found in dendritic spines and dendritic shafts at extrasynaptic and perisynaptic sites throughout postnatal development. We further demonstrated the presynaptic localization of GABABR1 and GABABR2, as well as the association of the receptors with asymmetrical synaptic junctions. These results indicate potentially important roles for the GABABRs in the regulation of migratory processes during corticogenesis and in the modulation of synaptic transmission during early development of cortical circuitry."}],"extern":"1","volume":15,"doi":"10.1046/j.1460-9568.2002.02032.x","day":"01","external_id":{"pmid":["12081656"]},"publisher":"Wiley-Blackwell","month":"06","date_published":"2002-06-01T00:00:00Z","article_processing_charge":"No","fulldoi":"https://doi.org/10.1046/j.1460-9568.2002.02032.x","publication_status":"published","year":"2002","quality_controlled":"1","pmid":1,"intvolume":" 15"},{"OA_type":"closed access","language":[{"iso":"eng"}],"publist_id":"4275","publication":"European Journal of Neuroscience","issue":"2","scopus_import":"1","publication_identifier":{"eissn":["1460-9568"],"issn":["0953-816X"]},"acknowledgement":"This work was supported by research grants from the Ministry of Education, Science, Sports and Culture of Japan, and the Japan Society for the Promotion of Science (P96319). We used subtype-specific antibodies in pre- and postembedding immunohistochemistry combined with three-dimensional reconstruction of labelled profiles and quantification of immunoparticles to reveal the subcellular distribution of pre- and postsynaptic GABABR1a/b and GABABR2 in the rat cerebellum and ventrobasal thalamus. GABABR1a/b and R2 were extensively colocalized in most brain regions including the cerebellum and thalamus. In the cerebellum, immunoreactivity for both subtypes was prevalent in the molecular layer. The most intense immunoreactivity was found in Purkinje cell spines with a high density of immunoparticles at extrasynaptic sites peaking at around 240 nm from glutamatergic synapses between spines and parallel fibre varicosities. This is in contrast to dendrites at sites around GABAergic synapses where sparse and random distribution was found for both subtypes. In addition, more than one-tenth of the synaptic membrane specialization of spine-parallel fibre synapses were labelled at pre- or postsynaptic sites. Weak immunolabelling for both subtypes was also seen in parallel fibres but only rarely in GABAergic axons. In the ventrobasal thalamus, immunolabelling for both receptor subtypes was intense over the dendritic field of thalamocortical cells. Electron microscopy demonstrated an extrasynaptic localization of GABABR1a/b and R2 exclusively in postsynaptic elements. Quantitative analysis further revealed the density of GABABR1a/b around GABAergic synapses was higher than glutamatergic synapses on thalamocortical cell dendrites. The distinct localization of GABABRs relative to synaptic sites in the cerebellum and ventrobasal thalamus suggests that GABABRs differentially regulate activity of different neuronal populations.","lang":"eng"}],"extern":"1","volume":15,"doi":"10.1046/j.0953-816x.2001.01855.x","day":"01","external_id":{"pmid":["11849296"]},"month":"01","publisher":"Wiley","date_published":"2002-01-01T00:00:00Z","article_processing_charge":"No","fulldoi":"https://doi.org/10.1046/j.0953-816x.2001.01855.x","quality_controlled":"1","pmid":1,"year":"2002","publication_status":"published","intvolume":" 15"},{"volume":13,"doi":"10.1046/j.0953-816X.2001.01494.x","external_id":{"pmid":["11285012"]},"month":"03","publisher":"Wiley-Blackwell","day":"01","fulldoi":"https://doi.org/10.1046/j.0953-816X.2001.01494.x","date_published":"2001-03-01T00:00:00Z","article_processing_charge":"No","quality_controlled":"1","year":"2001","pmid":1,"publication_status":"published","intvolume":" 13","publist_id":"4293","language":[{"iso":"eng"}],"publication_identifier":{"issn":["0953-816X"]},"acknowledgement":"MV and AF are senior coauthors of this work, which was supported by Ministerio de Educacion y Cultura, grants SAF97/0195 and SAF 2000-0152-C02-02 to M.V; PB94-0219-CO2-01 and PB97-0582-CO2-01 to A.F., Accio Especial de R+D AE98-18 from Generalitat Valenciana, and a Fellowship from Bancaixa-C.S.I.C. to J.R.M.-G. By using cortical slices of early postnatal mice, we show that in layer I cells, glutamate produces intracellular calcium ([Ca2+]i) elevations mediated by ionotropic and metabotropic glutamate receptors (mGluRs). The contribution of mGluRs to these responses was demonstrated by application of tACPD, an agonist to groups I and II mGluRs, which evoked [Ca2+]i increases that could be reversibly blocked by MCPG, an antagonist to groups I and II mGluRs. In the absence of extracellular Ca2+, repetitive applications of tACPD or quisqualate, an agonist to group I mGluRs, elicited decreasing [Ca2+]i responses that were restored by refilling a thapsigargin-sensitive Ca2+ store. The use of specific group I mGluR agonists CHPG and DHPG indicated that the functional mGluR in layer I was of the mGluR1 subtype. Pyramidal cells are thought to be phasically activated by spatial cues and a variety of sensory and motor stimuli. Here, we used a behavioural `space clamp' method, which involved the confinement of the actively running animal in a defined position in space (running wheel) and kept sensory inputs constant. Twelve percent of the recorded CA1 pyramidal cells were selectively active while the rat was running in the wheel. Cell firing was specific to the direction of running and disappeared after rotating the recording apparatus. The discharge frequency of pyramidal cells and interneurons was sustained as long as the rat ran continuously in the wheel. Furthermore, the discharge frequency of pyramidal cells and interneurons increased with increasing running velocity, even though the frequency of hippocampal theta waves remained constant. The discharge frequency of some `wheel-related' pyramidal cells could increase more than 10-fold between 10 and 100 cm/s, whereas the firing rate of `non-wheel' cells remained constantly low. We hypothesize that: (i) a necessary condition for place-specific discharge of hippocampal pyramidal cells is the presence of theta oscillation; and (ii) relevant stimuli can tonically and selectively activate hippocampal pyramidal cells as long as theta activity is present.","lang":"eng"}],"extern":"1","article_type":"original","type":"journal_article","oa_version":"None","citation":{"apa":"Czurkó, A., Hirase, H., Csicsvari, J. Here, we used a behavioural `space clamp' method, which involved the confinement of the actively running animal in a defined position in space (running wheel) to examine how `spatial' and other inputs affect firing rate and timing of hippocampal CA1 pyramidal cells and interneurons. Nineteen per cent of the recorded CA1 pyramidal cells were selectively active while the rat was running in the wheel in a given direction ('wheel' cells). Spatial rotation of the apparatus showed that selective discharge of pyramidal cells in the wheel was under the combined influence of distal and apparatus cues. During steady running, both discharge rate and theta phase were constant. Rotation of the wheel apparatus resulted in a shift of both firing rate and preferred theta phase. The discharge frequency of `wheel' cells increased threefold (on average) with increasing running velocity. In contrast, change in running speed had relatively little effect on the theta phase-related discharge of `wheel' cells. Our findings indicate that mechanisms that regulate rate and phase of spikes are overlapping but not necessarily identical."}],"extern":"1"},{"doi":"10.1111/j.1460-9568.1996.tb01611.x","volume":8,"day":"01","month":"07","publisher":"Wiley-Blackwell","external_id":{"pmid":["8758956 "]},"article_processing_charge":"No","date_published":"1996-07-01T00:00:00Z","fulldoi":"https://doi.org/10.1111/j.1460-9568.1996.tb01611.x","intvolume":" 8","quality_controlled":"1","year":"1996","pmid":1,"publication_status":"published","language":[{"iso":"eng"}],"publist_id":"4324","issue":"7","scopus_import":"1","publication":"European Journal of Neuroscience","publication_identifier":{"issn":["0953-816X"]},"citation":{"chicago":"Luján, Rafael, Zoltán Nusser, John Roberts, Ryuichi Shigemoto, and Péter Somogyi. “ Perisynaptic Location of Metabotropic Glutamate Receptors MGluR1 and MGluR5 on Dendrites and Dendritic Spines in the Rat Hippocampus.” European Journal of Neuroscience. In order to establish whether this pattern of distribution applies to the hippocampal principal cells and to other postsynaptic metabotropic glutamate receptors, the mGluR1a/b/c and mGluR5 subtypes were localized by immunocytochemistry. Principal cells in all hippocampal fields were reactive for mGluR5, the strata oriens and radiatum of the CA1 area being most strongly immunolabelled. Labelling for mGluR1b/c was strongest on some pyramids in the CA3 area, weaker on granule cells and absent on CA1 pyramids. Subpopulations of non-principal cells showed strong mGluR1 or mGluR5 immunoreactivity. Electron microscopic pre-embedding immunoperoxidase and both pre- and postembedding immunogold methods consistently revealed the extrasynaptic location of both mGluRs in the somatic and dendritic membrane of pyramidal and granule cells. The density of immunolabelling was highest on dendritic spines. At synapses, immunoparticles for both mGluR1 and mGluR5 were found always outside the postsynaptic membrane specializations. Receptors were particularly concentrated in a perisynaptic annulus around type 1 synaptic junctions, including the invaginations at 'perforated' synapses. Measurements of immunolabelling on dendritic spines showed decreasing levels of receptor as a function of distance from the edge of the synaptic specialization. We propose that glutamatergic synapses with an irregular edge develop in order to increase the circumference of synaptic junctions leading to an increase in the metabotropic to ionotropic glutamate receptor ratio at glutamate release sites. The perisynaptic position of postsynaptic metabotropic glutamate receptors appears to be a general feature of glutamatergic synaptic organization and may apply to other G-protein-coupled receptors. © European Neuroscience Association."}],"article_type":"original","oa_version":"None","type":"journal_article"},{"intvolume":" 5","year":"1993","quality_controlled":"1","pmid":1,"publication_status":"published","article_processing_charge":"No","date_published":"1993-11-01T00:00:00Z","main_file_link":[{"url":"https://onlinelibrary.wiley.com/doi/10.1111/j.1460-9568.1993.tb00213.x"}],"fulldoi":"https://doi.org/10.1111/j.1460-9568.1993.tb00213.x","day":"01","month":"11","publisher":"Wiley-Blackwell","external_id":{"pmid":["7904521 "]},"doi":"10.1111/j.1460-9568.1993.tb00213.x","volume":5,"date_created":"2018-12-11T12:07:24Z","author":[{"first_name":"Dan","last_name":"Lindholm","full_name":"Lindholm, Dan"},{"full_name":"Dechant, Georg","first_name":"Georg","last_name":"Dechant"},{"id":"39427864-F248-11E8-B48F-1D18A9856A87","first_name":"Carl-Philipp J","last_name":"Heisenberg","full_name":"Heisenberg, Carl-Philipp J","orcid":"0000-0002-0912-4566"},{"full_name":"Thoenen, Hans","last_name":"Thoenen","first_name":"Hans"}],"status":"public","_id":"4175","title":"Brain-derived neurotrophic factor is a survival factor for cultured rat cerebellar granule neurons and protects them against glutamate-induced neurotoxicity","abstract":[{"text":"We have studied the effects of different neurotrophins on the survival and proliferation of rat cerebellar granule cells in culture. These neurons express trkB and trkC, the putative neuronal receptors for brain-derived neurotrophic factor (BDNF) and neurotrophin-3 (NT-3) respectively. Binding studies using iodinated BDNF and NT-3 demonstrated that both BDNF and NT-3 bind to the cerebellar granule neurons with a similar affinity of approximately 2 x 10(-9) M. The number of receptors per granule cell was surprisingly high, approximately 30 x 10(-4) and 2 x 10(5) for BDNF and NT-3, respectively. Both NT-3 and BDNF elevated c-fos mRNA in the granule neurons, but only BDNF up-regulated the mRNA encoding the low-affinity neurotrophin receptor (p75). In contrast to NT-3, BDNF acted as a survival factor for the granule neurons. BDNF also induced sprouting of the granule neurons and significantly protected them against neurotoxicity induced by high (1 mM) glutamate concentrations. Cultured granule neurons also expressed low levels of BDNF mRNA which were increased by kainic acid, a glutamate receptor agonist. Thus, BDNF, but not NT-3, is a survival factor for cultured cerebellar granule neurons and activation of glutamate receptor(s) up-regulates BDNF expression in these cells.","lang":"eng"}],"extern":"1","article_type":"original","type":"journal_article","oa_version":"None","page":"1455 - 1464","citation":{"chicago":"Lindholm, Dan, Georg Dechant, Carl-Philipp J Heisenberg, and Hans Thoenen. “Brain-Derived Neurotrophic Factor Is a Survival Factor for Cultured Rat Cerebellar Granule Neurons and Protects Them against Glutamate-Induced Neurotoxicity.” European Journal of Neuroscience.
