text: 'With this status report, we aim to provide a timely snapshot of the protein
engineering field as a broad and rapidly advancing discipline that integrates
computational, molecular biology, structure-guided, evolutionary, and synthetic
approaches to create new and improved proteins with tailored structures and useful
functions. The report is organized into eight thematic areas spanning core methodologies
and major application domains, including enzymes, therapeutics, detection, synthetic
biology, and materials. Contributions from experts across these areas highlight
both the historical foundations and recent advances in their respective fields,
with particular emphasis on the growing influence of machine learning and artificial
intelligence-based methods. Emerging from this broad overview is a central message:
protein engineering appears to be entering a golden age, defined by a rapidly
accelerating pace of progress, even as significant challenges in design, screening,
and real-world application remain. Looking ahead, the continued integration of
computational and experimental strategies is poised to further accelerate the
impact of protein engineering across an expanding range of economically and societally
important sectors, from therapeutics and molecular imaging to diagnostics, plastic
    recycling, and industrial chemistry.'

text: We report the development of a continuous flow approach to nitrogen atom insertion.
The setup is modular, enabling the rapid optimization of reaction conditions for
a wide range of substrates. The milder reaction conditions led to an improved
substrate scope and functional group tolerance compared to batch conditions. The
reactions can also be safely scaled up to preparative scale.

text: Chemistry education at the graduate level and beyond faces the formidable
challenge of a boundless and constantly expanding frontier of knowledge on many
fronts. While modern learners have an increasingly broad range of resources available
at their disposal (including open access text-based references, online videos,
training problems, and other digital learning materials), there are comparatively
fewer such materials aimed at the highest levels of study. With the goal of producing
widely accessible graduate-level learning content, we created a community-based
approach to online course design that is easily digestible to meet the expectations
of modern learners. Herein, we report the development of an open access Advanced
Organic Chemistry video-based online course and several other specialized minicourses
using the Synthesis Workshop YouTube channel.

text: Biocatalysis has the potential to address the need for more sustainable organic
synthesis routes. Protein engineering can tune enzymes to perform in cascade reactions
and for efficient synthesis of enantiomerically enriched compounds, using both
natural and new-to-nature reaction pathways. This review highlights recent achievements
in biocatalysis, especially the development of novel enzymatic syntheses to access
versatile small molecule intermediates and complex biomolecules. Biocatalytic
strategies for the degradation of persistent pollutants and approaches for biomass
valorization are also discussed. The transition of chemical synthesis to a greener
future will be accelerated by implementing enzymes and engineering them for high
    performance and new activities.

text: Recent advances in single-atom insertion reactions have opened up new synthetic
approaches for molecular diversification. Developing innovative strategies to
directly transform biologically relevant molecules, without any prefunctionalization,
is key to further expanding the scope and utility of such transformations. Herein,
the direct access to quinazolines and pyrimidines from the corresponding unprotected
1H-indoles and 1H-pyrroles is reported, relying on the implementation of lithium
bis(trimethylsilyl)amide (LiHMDS) as a novel nitrogen atom source in combination
with commercially available hypervalent iodine reagents. Further application of
this strategy in late-stage settings demonstrates its potential in lead structure
    diversification campaigns.

text: The implementation of HCN-free transfer hydrocyanation reactions on laboratory
scales has recently been achieved by using HCN donor reagents under nickel- and
Lewis acid co-catalysis. More recently, malononitrile-based HCN donor reagents
were shown to undergo the C(sp3)–CN bond activation by the nickel catalyst in
the absence of Lewis acids. However, there is a lack of detailed mechanistic understanding
of the challenging C(sp3)–CN bond cleavage step. In this work, in-depth kinetic
and computational studies using alkynes as substrates were used to elucidate the
overall reaction mechanism of this transfer hydrocyanation, with a particular
focus on the activation of the C(sp3)–CN bond to generate the active H–Ni–CN transfer
hydrocyanation catalyst. Comparisons of experimentally and computationally derived
13C kinetic isotope effect data support a direct oxidative addition mechanism
of the nickel catalyst into the C(sp3)–CN bond facilitated by the coordination
of the second nitrile group to the nickel catalyst.

text: We report a convenient protocol for a nitrogen atom insertion into indenes
to afford isoquinolines. The reaction uses a combination of commercially available
phenyliodine(III) diacetate (PIDA) and ammonium carbamate as the nitrogen source
to furnish a wide range of isoquinolines. Various substitution patterns and commonly
used functional groups are well tolerated. The operational simplicity renders
this protocol broadly applicable and has been successfully extended towards the
direct interconversion of cyclopentadienes into the corresponding pyridines. Furthermore,
this strategy enables the facile synthesis of 15N labelled isoquinolines, using
    15NH4Cl as a commercial 15N source.

text: "A metal-free deaminative coupling of non-prefunctionalised benzylamines and
arylboronic acids is reported. In this operationally simple reaction, a primary
amine in benzylamine is converted into a good leaving group in situ using inexpensive
and commercially available isoamyl nitrite as a nitrosating reagent. Lewis-acidic
arylboronic acids are shown to replace mineral acids such as HCl or HBF4 that

text: Compared with peripheral late-stage transformations mainly focusing on carbon–hydrogen
functionalizations, reliable strategies to directly edit the core skeleton of
pharmaceutical lead compounds still remain scarce despite the recent flurry of
activity in this area. Herein, we report the skeletal editing of indoles through
nitrogen atom insertion, accessing the corresponding quinazoline or quinoxaline
bioisosteres by trapping of an electrophilic nitrene species generated from ammonium
carbamate and hypervalent iodine. This reactivity relies on the strategic use
of a silyl group as a labile protecting group that can facilitate subsequent product
release. The utility of this highly functional group-compatible methodology in
the context of late-stage skeletal editing of several commercial drugs is demonstrated.

text: Hydrocyanation reactions enable access to synthetically valuable nitriles
from readily available alkene precursors. However, hydrocyanation reactions using
hydrogen cyanide (HCN) or similarly toxic reagents on laboratory scale can be
particularly challenging due to their hazardous nature. In addition, such processes
typically require air- and temperature-sensitive Ni(0) precatalysts, further reducing
the operational simplicity of this transformation. Herein, we report a HCN-free
transfer hydrocyanation of alkenes and alkynes that employs commercially available
aliphatic nitriles as sacrificial HCN donors in combination with a catalytic amount
of air-stable and inexpensive NiCl2 as a precatalyst and a cocatalytic Lewis acid.
The scalability and robustness of the catalytic process were demonstrated by the
hydrocyanation of α-methylstyrene on a 100 mmol scale (11.4 g of product obtained)
using 1 mol % of the Ni catalyst. In addition, the feasibility of the dehydrocyanation
protocol using the air-stable Ni(II) precatalyst and norbornadiene as a sacrificial
acceptor was showcased by the selective conversion of an aliphatic nitrile into
    the corresponding alkene.

text: Reversible catalytic reactions operate under thermodynamic control, and thus,
establishing a selective catalytic system poses a considerable challenge. Herein,
we report a reversible transfer hydrocyanation protocol that exhibits high selectivity
for the thermodynamically less favorable branched isomer. Selectivity is achieved
by exploiting the lower barrier for C–CN oxidative addition and reductive elimination
at benzylic positions in the absence of a cocatalytic Lewis acid. Through the
design of a novel type of HCN donor, a practical, branched-selective, HCN-free
transfer hydrocyanation was realized. The synthetically useful resolution of a
mixture of branched and linear nitrile isomers was also demonstrated to underline
the value of reversible and selective transfer reactions. In a broader context,
this work demonstrates that high kinetic selectivity can be achieved in reversible
transfer reactions, thus opening new horizons for their synthetic applications.
