text: "Instruction-tuned Large Language Models (LLMs) show impressive results in
numerous practical applications, but they lack essential safety features that
are common in other areas of computer science, particularly an explicit separation
of instructions and data. This makes them vulnerable to manipulations such as
indirect prompt injections and generally unsuitable for safety-critical tasks.
Surprisingly, there is currently no established definition or benchmark to quantify
this phenomenon. In this work, we close this gap by introducing a formal measure
for instruction-data separation and an empirical variant that is calculable from
a model's outputs. We also present a new dataset, SEP, that allows estimating
the measure for real-world models. Our results on various LLMs show that the problem
of instruction-data separation is real: all models fail to achieve high separation,
and canonical mitigation techniques, such as prompt engineering and fine-tuning,
either fail to substantially improve separation or reduce model utility. The source
code and SEP dataset are openly accessible at https://github.com/egozverev/Shold-It-Be-Executed-Or-Processed.\r\n"

text: Dynamic processes in molecules can occur on a wide range of timescales, and
it is important to understand which timescales of motion contribute to different
parameters used in dynamics measurements. For spin relaxation, this can easily
be understood from the sampling frequencies of the spectral-density function by
different relaxation-rate constants. In addition to data from relaxation measurements,
determining dynamically averaged anisotropic interactions in magic-angle spinning
(MAS) solid-state NMR allows for better quantification of the amplitude of molecular
motion. For partially averaged anisotropic interactions, the relevant timescales
of motion are not so clearly defined. Whether the averaging depends on the experimental
methods (e.g., pulse sequences) or conditions (e.g., MAS frequency, magnitude
of anisotropic interaction, radio-frequency field amplitudes) is not fully understood.
To investigate these questions, we performed numerical simulations of dynamic
systems based on the stochastic Liouville equation using several experiments for
recoupling the dipolar coupling, chemical-shift anisotropy or quadrupolar coupling.
As described in the literature, the transition between slow motion, where parameters
characterizing the anisotropic interaction are not averaged, and fast motion,
where the tensors are averaged leading to a scaled anisotropic quantity, occurs
over a window of motional rate constants that depends mainly on the strength of
the interaction. This transition region can span 2 orders of magnitude in exchange-rate
constants (typically in the microsecond range) but depends only marginally on
the employed recoupling scheme or sample spinning frequency. The transition region
often coincides with a fast relaxation of coherences, making precise quantitative
measurements difficult. Residual couplings in off-magic-angle experiments, however,
average over longer timescales of motion. While in principle one may gain information
on the timescales of motion from the transition area, extracting such information
is hampered by low signal-to-noise ratio in experimental spectra due to fast relaxation
that occurs in the same region.

text: It is widely believed that information storage in neuronal circuits involves
nanoscopic structural changes at synapses, resulting in the formation of synaptic
engrams. However, direct evidence for this hypothesis is lacking. To test this
conjecture, we combined chemical potentiation, functional analysis by paired pre-postsynaptic
recordings, and structural analysis by electron microscopy (EM) and freeze-fracture
replica labeling (FRL) at the rodent hippocampal mossy fiber synapse, a key synapse
in the trisynaptic circuit of the hippocampus. Biophysical analysis of synaptic
transmission revealed that forskolin-induced chemical potentiation increased the
readily releasable vesicle pool size and vesicular release probability by 146%
and 49%, respectively. Structural analysis of mossy fiber synapses by EM and FRL
demonstrated an increase in the number of vesicles close to the plasma membrane
and the number of clusters of the priming protein Munc13-1, indicating an increase
in the number of both docked and primed vesicles. Furthermore, FRL analysis revealed
a significant reduction of the distance between Munc13-1 and CaV2.1 Ca2+ channels,
suggesting reconfiguration of the channel-vesicle coupling nanotopography. Our
results indicate that presynaptic plasticity is associated with structural reorganization
of active zones. We propose that changes in potential nanoscopic organization
at synaptic vesicle release sites may be correlates of learning and memory at
    a plastic central synapse.

text: It is widely believed that information storage in neuronal circuits involves
nanoscopic structural changes at synapses, resulting in the formation of synaptic
engrams. However, direct evidence for this hypothesis is lacking. To test this
conjecture, we combined chemical potentiation, functional analysis by paired pre-postsynaptic
recordings, and structural analysis by electron microscopy (EM) and freeze-fracture
replica labeling (FRL) at the murine hippocampal mossy fiber synapse, a key synapse
in the trisynaptic circuit of the hippocampus.
We propose that changes in potential nanoscopic organization
at synaptic vesicle release sites may be correlates of learning and memory at
    a plastic central synapse.

text: 'Surveys in the Milky Way and Large Magellanic Cloud have revealed that the
majority of massive stars will interact with companions during their lives. However,
knowledge of the binary properties of massive stars at low metallicity, and therefore
in conditions approaching those of the Early Universe, remain sparse. We present
the Binarity at LOw Metallicity (BLOeM) campaign, an ESO large programme designed
to obtain 25 epochs of spectroscopy for 929 massive stars in the Small Magellanic
Cloud, allowing us to probe multiplicity in the lowest-metallicity conditions
to date (Z = 0.2 Z⊙). BLOeM will provide (i) the binary fraction, (ii) the orbital
configurations of systems with periods of P ≲ 3 yr, (iii) dormant black-hole binary
candidates (OB+BH), and (iv) a legacy database of physical parameters of massive
stars at low metallicity. Main sequence (OB-type) and evolved (OBAF-type) massive
stars are observed with the LR02 setup of the GIRAFFE instrument of the Very Large
    Telescope (3960–4570 Å resolving power R = 6200; typical signal-to-noise ratio(S/N)
≈70–100). This paper utilises the first nine epochs obtained over a three-month
time period. We describe the survey and data reduction, perform a spectral classification
of the stacked spectra, and construct a Hertzsprung-Russell diagram of the sample
via spectral-type and photometric calibrations. Our detailed classification reveals
that the sample covers spectral types from O4 to F5, spanning the effective temperature
    and luminosity ranges 6.5 ≲ Teff/kK ≲ 45 and 3.7 < log L/L⊙ < 6.1 and initial
    masses of 8 ≲ Mini ≲ 80 M⊙. The sample comprises 159 O-type stars, 331 early B-type
(B0–3) dwarfs and giants (luminosity classes V–III), 303 early B-type supergiants
(II–I), and 136 late-type BAF supergiants. At least 82 stars are OBe stars: 20
O-type and 62 B-type (13% and 11% of the respective samples). In addition, the
sample includes 4 high-mass X-ray binaries, 3 stars resembling luminous blue variables,
2 bloated stripped-star candidates, 2 candidate magnetic stars, and 74 eclipsing
    binaries.'

text: Precision sensing and manipulation of milligram-scale mechanical oscillators
has attracted growing interest in the fields of table-top explorations of gravity
and tests of quantum mechanics at macroscopic scales. Torsional oscillators present
an opportunity in this regard due to their remarked isolation from environmental
noise. For torsional motion, an effective employment of optical cavities to enhance
optomechanical interactions—as already established for linear oscillators—so far
faced certain challenges. Here, we propose a concept for sensing and manipulating
torsional motion, where exclusively the torsional rotations of a pendulum are
mapped onto the path length of a single two-mirror optical cavity. The concept
inherently alleviates many limitations of previous approaches. A proof-of-principle
experiment is conducted with a rigidly controlled pendulum to explore the sensing
aspects of the concept and to identify practical limitations in a potential state-of-the
art setup. Based on this study, we anticipate development of precision torque
sensors utilizing torsional pendulums that can support sensitivities below 10−19Nm/√Hz,
while the motion of the pendulums are dominated by quantum radiation pressure
noise at sub-microwatts of incoming laser power. These developments will provide
horizons for experiments at the interface of quantum mechanics and gravity.

text: Proteins often undergo large-scale conformational transitions, in which secondary
and tertiary structure elements (loops, helices, and domains) change their structures
or their positions with respect to each other. Simple considerations suggest that
such dynamics should be relatively fast, but the functional cycles of many proteins
are often relatively slow. Sophisticated experimental methods are starting to
tackle this dichotomy and shed light on the contribution of large-scale conformational
dynamics to protein function. In this review, we focus on the contribution of
single-molecule Förster resonance energy transfer and nuclear magnetic resonance
(NMR) spectroscopies to the study of conformational dynamics. We briefly describe
the state of the art in each of these techniques and then point out their similarities
and differences, as well as the relative strengths and weaknesses of each. Several
case studies, in which the connection between fast conformational dynamics and
slower function has been demonstrated, are then introduced and discussed. These
examples include both enzymes and large protein machines, some of which have been
studied by both NMR and fluorescence spectroscopies.

text: 'The hippocampal mossy fiber synapse, formed between axons of dentate gyrus
granule cells and dendrites of CA3 pyramidal neurons, is a key synapse in the
trisynaptic circuitry of the hippocampus. Because of its comparatively large size,
this synapse is accessible to direct presynaptic recording, allowing a rigorous
investigation of the biophysical mechanisms of synaptic transmission and plasticity.
Furthermore, because of its placement in the very center of the hippocampal memory
circuit, this synapse seems to be critically involved in several higher network
functions, such as learning, memory, pattern separation, and pattern completion.
Recent work based on new technologies in both nanoanatomy and nanophysiology,
including presynaptic patch-clamp recording, paired recording, super-resolution
light microscopy, and freeze-fracture and “flash-and-freeze” electron microscopy,
has provided new insights into the structure, biophysics, and network function
of this intriguing synapse. This brings us one step closer to answering a fundamental
question in neuroscience: how basic synaptic properties shape higher network computations.'

text: DNA cloning is a core technique in biomedical and biotechnological research
and is used to assemble and modify DNA fragments at will. While DNA cloning has
traditionally relied on restriction enzymes, recent homology-based methods offer
improved protocols together with seamless and directional assembly of desired
products, overcoming the main disadvantages of restriction enzyme DNA cloning.
This chapter provides a historical perspective on DNA cloning, presents a detailed
discussion on state-of-the-art in vitro and in vivo homology-based methodologies,
covering the basics of how to perform all major plasmid modifications (sub-cloning,
site-directed mutagenesis, insertions, and deletions), and gives examples of how
to apply these techniques for complex DNA cloning projects.

text: Many scientific breakthroughs have depended on animal research, yet the ethical
concerns surrounding the use of animals in experimentation have long prompted
discussions about humane treatment and responsible scientific practice. First
articulated by Russell and Burch, the 3Rs Principles of Replacement, Reduction,
and Refinement have gained widespread recognition as basic guidelines for animal
research. Over time, the 3Rs have transcended the research community, influencing
policy decisions, animal welfare advocacy and public perception of animal experimentation.
Despite their broad acceptance, interpretations of the 3Rs vary substantially,
shaping statutory frameworks at various levels, with both technical and practical
    impacts.

text: Light-driven rotary molecular motors are among the most promising classes
of responsive molecular machines and take advantage of their intrinsic chirality
which governs unidirectional rotation. As a consequence of their dynamic function,
they receive considerable interest in the areas of supramolecular chemistry, asymmetric
catalysis and responsive materials. Among the emerging classes of responsive photochromic
molecules, multistate first-generation molecular motors driven by benign visible
light remain unexplored, which limits the exploitation of the full potential of
these mechanical light-powered systems. Herein, we describe a series of all-visible-light-driven
first-generation molecular motors based on the salicylidene Schiff base functionality.
Remarkable redshifts up to 100 nm in absorption are achieved compared to conventional
first-generation motor structures. Taking advantage of all-visible-light-driven
multistate motor scaffolds, adaptive behaviour is found as well, and potential
application in multistate photoluminescence is demonstrated. These functional
visible-light-responsive motors will likely stimulate the design and synthesis
of more sophisticated nanomachinery with a myriad of future applications in powering
    dynamic systems.

text: The development of photoresponsive systems with non-invasive orthogonal control
by distinct wavelengths of light is still in its infancy. In particular, the design
of photochemically triggered-orthogonal systems integrated into solid materials
that enable multiple dynamic control over their properties remains a longstanding
challenge. Here, we report the orthogonal and reversible control of two types
of photoswitches in an integrated solid porous framework, that is, visible-light
responsive o-fluoroazobenzene and nitro-spiropyran motifs. The properties of the
constructed material can be selectively controlled by different wavelengths of
light thus generating four distinct states providing a basis for dynamic multifunctional
materials. Solid-state NMR spectroscopy demonstrated the selective transformation
of the azobenzene switch in the bulk, which in turn modulates N2 and CO2 adsorption.

text: Photoisomerization and photoluminescence are two distinct energy dissipation
pathways in light-driven molecular motors. The photoisomerization properties of
discrete molecular motors have been well established in solution, but their photoluminescent
properties have been rarely reported—especially in aggregates. Here, it is shown
that an overcrowded alkene-based molecular motor exhibits distinct dynamic properties
in solution and aggregate states, for example, gel and solid states. Despite the
poor emissive properties of molecular motors in solution, a bright emission is
observed in the aggregate states, including in gel and the crystalline solid.
The emission wavelength is highly dependent on the nature of the supramolecular
packing and order in the aggregates. As a result, the fluorescent color can be
readily tuned reversibly via mechanical grinding and vapor fuming, which provides
a new platform for developing multi-stimuli functional materials.

text: We consider a gas of N weakly interacting bosons in the ground state. Such
gases exhibit Bose–Einstein condensation. The binding energy is defined as the
energy it takes to remove one particle from the gas. In this article, we prove
an asymptotic expansion for the binding energy, and compute the first orders explicitly
for the homogeneous gas. Our result addresses in particular a conjecture by Nam
(Lett Math Phys 108(1):141–159, 2018), and provides an asymptotic expansion of
the ionization energy of bosonic atoms.

text: "We study the Fröhlich polaron model in R3, and prove a lower bound on its
ground state energy as a function of the total momentum. The bound is asymptotically
sharp at large coupling. In combination with a corresponding upper bound proved
earlier (Mitrouskas et al. in Forum Math. Sigma 11:1–52, 2023), it shows that
the energy is approximately parabolic below the continuum threshold, and that
the polaron’s effective mass (defined as the semi-latus rectum of the\r\nparabola)
is given by the celebrated Landau–Pekar formula. In particular, it diverges as
α4 for large coupling constant α."

text: We consider a gas of N bosons with interactions in the mean-field scaling
regime. We review a recent proof of the asymptotic expansion of its spectrum and
eigenstates and two applications of this result, namely the derivation of an Edgeworth
expansion for fluctuations of one-body operators and the computation of the binding
energy of an inhomogeneous Bose gas to any order. Finally, we collect related
results for the dynamics of the weakly interacting Bose gas and for the regularized
    Nelson model.

text: "Autoantibodies against the protein leucine-rich glioma inactivated 1 (LGI1)
cause the most\r\ncommon subtype of autoimmune encephalitis with predominant involvement
of the limbic\r\nsystem, associated with seizures and memory deficits. LGI1 and
its receptor ADAM22 are part\r\nof a transsynaptic protein complex that includes
several proteins involved in presynaptic\r\nneurotransmitter release and postsynaptic
glutamate sensing. Autoantibodies against LGI1\r\nincrease excitatory synaptic
strength, but studies that genetically disrupt the LGI1-ADAM22\r\ncomplex report
a reduction in postsynaptic glutamate receptor-mediated responses. Thus, the\r\nmechanisms
underlying the increased synaptic strength induced by LGI1 autoantibodies remain
elusive, and the contributions of presynaptic molecules to the LGI1-transsynaptic
complex remain unclear. We therefore investigated the presynaptic mechanisms that
    mediate\r\nautoantibody-induced synaptic strengthening."

text: This workshop brought together experts on the analysis of quantum many-body
problems and quantum statistical mechanics, with the goal of discussing the state-of-the-art
of the field, recent developments as well as challenges for the future. The main
topics of discussion concerned the equilibrium and dynamical behavior of (bosonic
or fermionic) quantum gases, quantum spin systems, as well as quantum field theory
models like the Nelson or Fröhlich model.
acknowledgement: The MFO and the workshop organizers would like to thank the National
Science Foundation for supporting the participation of junior researchers in the
workshop by the grant DMS-2230648, “US Junior Oberwolfach Fellows”.
